タイトル; Efficient transport and storage system for genetically engineered mice using refrigeration and cryopreservation of sperm.

要旨;
Due to the spread of the new coronavirus, breeding staff and researchers have restricted to enter animal facilities and had to reduce the number of laboratory mice. Cryopreservation of sperm and embryos in mouse banks has been effective in backing up valuable mice. However, it did not work in some facilities due to difficulties in transporting living mouse organisms during the corona disaster. On the other hand, in the CARD mouse bank, we have developed a technology to transport cold-stored mouse sperm as an alternative to biological transport. And we were able to cryopreserve the sperm and make fertilized eggs using the sperm in the CARD mouse bank using this technology. In the technique of cold-transport of mouse sperm, sperm preservation solution supplemented with dimethyl sulfoxide (DMSO) and quercetin was used to maintain the fertilization ability of cold-stored sperm for up to 10 days. Furthermore, the DMSO and quercetin concentrations were optimized to allow cryopreservation of cold-stored sperm. In this study, we will report data on the optimal concentrations of DMSO and quercetin for refrigeration and cryopreservation of sperm. In addition, we will show result on the refrigerated transport and cryopreservation of sperm from Japan and abroad using this technology. In summary, these findings are useful as an efficient transport and storage system for genetically engineered mice that can be used in the event of a corona pandemic.
