transcriptionally permissive heterochromatin at germline genes by assembling PRC1.6 and Compass-like complexes.

要旨;
Two states of pluripotency, called naive and primed, have been described in both rodents and humans. However, it is still not fully understood how these two pluripotent states are established and maintained. Germline genes are a representative group of genes differentially expressed in naive and primed pluripotent stem cells (PSCs). We previously reported that PRC1.6, an atypical Polycomb repressive complex 1 (PRC1), is responsible for silencing of germline genes and thereby contributes to self-renewal of mouse ES cells (mESCs; naive PSCs). We further found that naive pluripotency-specific transcription factors Dppa2 and 4 interact with PRC1.6 and Compass-like complexes, and assemble these complexes at germline genes in mESCs. In contrast, these complexes do not bind to germline genes in mouse epiblast stem cells (mEpiSCs; primed PSCs) in which Dppa2/4 are absent. We propose that Dppa2/4 establish naïve pluripotency-specific transcriptionally permissive heterochromatin at germline genes by assembling PRC1.6 and Compass-like complexes.
