We propose a divided recognition of the full-length target (27 mer, WT27) containing SNP (single nucleotide polymorphism) site through cooperative hybridization of two kinds of DNA conjugates. One of them is a 7 mer oligonucleotide (ODN) tethered with EDTA at 5' terminus (capture probe, C7). It is complementary to the 7 mer sequence in WT27 containing SNP site. The other is a 20 mer ODN conjugate bearing o-phenanthroline (phen) at 3' terminus (sensitizer probe, S20). The modified ligands, EDTA and phen, were designed to locate face to face on the target providing microenvironment to accommodate lanthanide ions (Tb3+ and Eu3+). In this system, the emissions from the lanthanide ions were clearly observed in the presence of WT27. However, the emissions were scarcely observed in the presence of Wut27 containing one base mismatch for C7. Both of the conjugates would cooperatively coordinate to a lanthanide ion to form luminous complex only with the guide of WT27.
