タイトル; E3enzyme RAD18 suppresses cellular senescence.

Various kinds of exogenous and endogenous DNA damaging agents cause stalling of replication, that is detrimental for the cellular growth. To overcome them, E3 ligase RAD18 play a crucial role for maintaining the genomic integrity through the mechanism called DNA damage tolerance. Unexpectedly, Rad18 knockout mouse showed premature aging phenotypes in some organs, such as testis. To reveal the impact of RAD18 on prevention of the aging phenotypes, we have compared extent of cellular senescence between WT and Rad18-/- MEFs under spontaneous or following UV irradiation conditions. Rad18-/- MEFs tended to be cellular senescence at higher frequency than those of WT MEFs in both conditions. We will also use human primary cells to examine the impact of RAD18 for extent of cellular senescence and expression of a cellular senescence marker, CDK inhibitor p16 under conditions of spontaneous or after UV irradiation. We would like to discuss how RAD18 suppresses cellular senescence in the conditions.
