タイトル; Development and improvement of cold-storage technology for rat sperm.

Transport technology of laboratory rats is an important topic in reproductive technology. Cold-transport of cauda epididymis provides a simple solution for transporting genetic resource in mice. Previously, we found dimethyl sulfoxide (DMSO) and quercetin prolonged the survival time of mouse sperm at refrigerated temperatures. However, the compatibility of the cold-storage technique using DMSO and quercetin was not examined in rat sperm. In this study, we examined the effects of DMSO and quercetin on the cold-storage of rat sperm. DMSO and quercetin maintained the motility and fertility of the cold-stored rat sperm up to 5 days. The rates of fertilization dropped time-dependently. In addition, we confirmed two-cell embryos derived from in vitro fertilization using the cold-stored rat sperm normally developed to pups by embryo transfer. Finally, rat sperm were transported using the cold-transport system from Niigata University to Kumamoto University and the sperm had fertilization and developmental abilities. In conclusion, this study suggests that DMSO and quercetin are effective in the cold storage of rat sperm. In the future, we will continue to improve the techniques of cold transport of rat sperm and in vitro fertilization to use for the international transportation.
